Vitamin B1 (Thiamine) Thiamine is the cofactor for three key enzymes in carbohydrate and branched-chain amino acid metabolism: pyruvate dehydrogenase, alpha-ketoglutarate dehydrogenase, and transketolase. The first two are essential steps in the Krebs cycle that produces ATP; the third is required for the pentose phosphate pathway that generates NADPH and ribose for nucleic acid synthesis. Vitamin B2 (Riboflavin) Riboflavin is the precursor for flavin adenine dinucleotide (FAD) and flavin mononucleotide (FMN) — two coenzymes that are central electron carriers in the mitochondrial electron transport chain. Without adequate FAD and FMN, the electron transport chain cannot transfer electrons efficiently to molecular oxygen to generate the proton gradient that drives ATP synthesis. Vitamin B3 (Niacin) Niacin is the precursor for NAD+ and NADP+ — the most important redox coenzymes in human metabolism. NAD+ participates in over 500 enzymatic reactions as an electron acceptor, including the Krebs cycle, beta-oxidation of fatty acids, and the glycolytic pathway. NADP+ is the electron donor in reductive biosynthesis — fatty acid synthesis, cholesterol synthesis, and the antioxidant regeneration of glutathione. At higher concentrations, niacin also directly modulates lipoprotein metabolism, reducing triglyceride production and LDL oxidation while supporting HDL synthesis.


Vitamin B5 (Pantothenic Acid) Pantothenic acid is the structural component of coenzyme A (CoA) — the universal acetyl group carrier in metabolism. CoA is required for acetyl-CoA production at the entry point of the Krebs cycle, for fatty acid synthesis and oxidation, for the synthesis of steroid hormones and bile acids, and for the acetylation of proteins in post-translational modification. Without B5, CoA cannot be synthesised and these metabolic pathways stall.


Vitamin B6 (Pyridoxine) Pyridoxal-5-phosphate (the active form of B6) is a cofactor in over 100 enzymatic reactions, more than any other B-vitamin. It is the essential cofactor for amino acid transamination — the process by which amino acids are interconverted and their nitrogen is transferred for excretion — and for the synthesis of neurotransmitters including dopamine, serotonin, GABA, and norepinephrine from their amino acid precursors. It is also required for haemoglobin synthesis and for glycogen phosphorylase, the enzyme that mobilizes glucose from glycogen stores during exercise.


Vitamin B9 (Folic Acid) Folate is the cofactor for one-carbon transfer reactions in the synthesis and methylation of DNA. It is required for the de novo synthesis of purines and thymidine — the nucleotide bases in DNA — making it essential for cell division in all rapidly proliferating tissues. Working alongside B12 in the methylation cycle, folate converts homocysteine to methionine, providing the methyl groups for DNA methylation that regulates gene expression. Folate's role in neural tube development makes it particularly critical in early pregnancy, but its requirement for DNA synthesis and repair is lifelong.


Vitamin B12 (Cyanocobalamin) B12 is the cofactor for methionine synthase in the folate-methionine methylation cycle and for methylmalonyl-CoA mutase in odd-chain fatty acid catabolism. Vitamin E (Tocopherol) Vitamin E is the primary fat-soluble antioxidant in cell membranes. Cell membranes are composed largely of polyunsaturated fatty acids, which are highly susceptible to lipid peroxidation by reactive oxygen species — a chain reaction that degrades membrane integrity and triggers inflammatory signalling. Vitamin E terminates this chain reaction by donating a hydrogen atom to the lipid peroxide radical, stopping peroxidation from propagating through the membrane.
